Abstract: The production of controlled bacterial cellulose structures for various applications requires a better understanding on the mechanism of cellulose biosynthesis as well as proper tools for structural characterization of the materials. In this work, bacterial celluloses synthesized by an Asaia bogorensis strain known to produce fine cellulose fibrils and a commonly used Komagataeibacter xylinus strain were characterized using a comprehensive set of methods covering multiple levels of the hierarchical structure. FT-IR spectroscopy and x-ray diffraction were used to analyse the crystal structure and crystallite dimensions, whereas scanning and transmission electron microscopy, atomic force microscopy, and small-angle x-ray and neutron scattering were employed to obtain information on the higher-level fibrillar structures. All methods yielded results consistent with the A. bogorensis cellulose fibrils being thinner than the K. xylinus fibrils on both the level of individual cellulose microfibrils and bundles or ribbons thereof, even though the exact values determined for the lateral fibril dimensions depended slightly on the method and sample preparation. Particularly, the width of microfibril bundles determined by the microscopy methods differed due to shrinkage and preferred orientation caused by drying, whereas the microfibril diameter remained unaffected. The results were used to understand the biological origin of the differences between the two bacterial celluloses.

Abstract: A scanning X-ray diffraction study of cardiac tissue has been performed, covering the entire cross section of a mouse heart slice. To this end, moderate focusing by compound refractive lenses to micrometer spot size, continuous scanning, data acquisition by a fast single-photon-counting pixel detector, and fully automated analysis scripts have been combined. It was shown that a surprising amount of structural data can be harvested from such a scan, evaluating the local scattering intensity, interfilament spacing of the muscle tissue, the filament orientation, and the degree of anisotropy. The workflow of data analysis is described and a data analysis toolbox with example data for general use is provided. Since many cardiomyopathies rely on the structural integrity of the sarcomere, the contractile unit of cardiac muscle cells, the present study can be easily extended to characterize tissue from a diseased heart.

Abstract: The application of IR spectroscopy to the characterization and quality control of samples used in neutron crystallography is described. While neutron crystallography is a growing field, the limited availability of neutron beamtime means that there may be a delay between crystallogenesis and data collection. Since essentially all neutron crystallographic work is carried out using Dsb 2O-based solvent buffers, a particular concern for these experiments is the possibility of Hsb 2O back-exchange across reservoir or capillary sealants. This may limit the quality of neutron scattering length density maps and of the associated analysis. Given the expense of central facility beamtime and the effort that goes into the production of suitably sized (usually perdeuterated) crystals, a systematic method of exploiting IR spectroscopy for the analysis of back-exchange phenomena in the reservoirs used for crystal growth is valuable. Examples are given in which the characterization of Dsb 2O/Hsb 2O back-exchange in transthyretin crystals is described.
